An In-situ Solvent Recycling Process for SPPS

The use of high efficiency solid phase peptide synthesis (HE-SPPS) provides benefits for the synthesis of peptides due to its high purity, rapid speed, and low chemical usage characteristcs [1].  This has been advanced further through the development of a process whereby the deprotection is added directly to the previous undrained coupling solution [2].  This was found possible based on the rapid kinetics of quenching between the deprotection base and activated amino acid which prevented potential side reactions.  As a separate improvement, this process was further enhanced for the advent of a new vacuum assisted base removal during the deprotection step at elevated temperature [3].  The effectiveness of these new processes were tested on the sequential synthesis of a batch of 24 peptides.
