Summary

The use of high efficiency solid phase peptide synthesis (HE-SPPS) allows synthesis of peptides at a previously unprecedented level of high speed, high purity, and low production of chemical waste. These developments allow the production of a difficult 10 mer peptide (0.1mmol scale) in < 30 minutes with < 100mL total waste. This technology can be readily extended to produce a 50 mer peptide in 3.5 hours. It has also permitted scale up into the kilogram range with ease.

In this presentation, the most recent advances in microwave assisted SPPS are reported. Topics include: (1) review of current methodology, (2) introduction of new methodology for GMP high-throughput synthesis, and (3) scale up from milligram to kilogram quantities of peptide. The robust nature of these methodologies is demonstrated with the high-purity syntheses of a number of pharmaceutically relevant peptides as well post-synthetically sequences.
